Is there a way I can make this work running Sierra? I just found out that their website says it works for Mac OS up to 10.11.
If it won't work with Sierra, is there a new alternative?

It looks like you need to install Keyspan 4.0 without disabling the SIP (System Integrity Protection) according to this article on Francis Fustier's blog:

Sorry that didn't work for you, Jack.
Before installing v4, Francis also recommends that you delete the older version of the Keyspan driver from:
• {your hard drive} /Library/Extensions
• {your hard drive} /System/Library/Extensions
and empty the trash.
If that doesn't work either, try disabling the SIP and then installing v4.

P.S.: I replaced my Keyspan some time ago with a Shipmodul Mutliplexer that has a direct USB cable or WiFi for the Mac.
